Development of chitosan, pullulan, and alginate based drug-loaded nano-emulsions as a potential malignant melanoma delivery platform

Melanoma is the most aggressive form of skin cancer and various treatments have been investigated to treat this disease, but drug resistance remains an important factor in failure conventional therapeutics. Here we describe development, optimisation characterisation alginate, chitosan, pullulan, their combined nano-emulsions as delivery platforms for potential application melanoma. A novel nano-emulsion system was designed assessed by determining vitro release, cell viability (MTT), cellular apoptosis (ELISA) confocal microscopy. comparative analysis effect on BRAF -mutant melanoma (A375) keratinocyte (HaCaT) cells conducted, with “pullulan-chitosan” chosen approach delivery. Increased induction recorded 90% after 72 hours treatment doxorubicin-loaded optimal nano-emulsion. Similarly, same treatment, decreased 70%. More importantly, A375 treated naïve doxorubicin were 100% viable compared which only 30%viable. Achieved results are indicating importance carrier's polymeric combination impact release pattern efficiency treatment. This offers abrogation drug-efflux-related chemo-resistance.
